Kymera Therapeutics Presents New Phase 1 Data for KT-621 in Healthy Japanese Adults at JDA Annual Meeting

(NASDAQ: KYMR), a clinical-stage biopharmaceutical company developing novel oral small molecule degrader medicines for immunological diseases, announced today that results from the Phase 1 study of its investigational drug KT-621, a first-in-class, oral STAT6 degrader, in healthy Japanese adults were presented at the Japanese Dermatological Association (JDA) Annual Meeting. The presentation shared safety and pharmacokinetic/pharmacodynamic (PK/PD) data consistent with previously reported KT-621 clinical studies.

KT-621 is designed to target STAT6, the transcription factor central to Type 2 inflammation driven by IL-4/IL-13 signaling. The company is currently conducting parallel Phase 2b trials for KT-621 in atopic dermatitis and asthma. Data from the atopic dermatitis trial, BROADEN2, is anticipated by mid-2027, and data from the asthma trial, BREADTH, is expected by late 2027. These Phase 2b studies aim to accelerate KT-621 development for subsequent Phase 3 registration studies across multiple Type 2 inflammatory diseases.

In the Phase 1 trial involving 24 healthy Japanese adults, KT-621 demonstrated a favorable PK profile, including rapid absorption and dose-proportional increases in plasma exposure. The drug also showed rapid and sustained STAT6 degradation in blood, with median degradation of 98% or higher at Day 7 across both dose levels tested. The study, which was randomized, double-blind, and placebo-controlled, administered KT-621 or placebo once daily for seven days at two dose levels in a 3:1 ratio. KT-621 was well tolerated and exhibited a favorable safety profile at both doses.

These findings in healthy Japanese adults were comparable to data previously observed in non-Japanese adults and patients with moderate to severe atopic dermatitis, further supporting KT-621’s potential as an oral treatment for chronic Type 2 inflammatory diseases. The Phase 1 trial in Japan was designed to gather the necessary PK/PD and safety data required by regulators before initiating patient enrollment in Japan for the global Phase 2b studies.

Previous Phase 1 data for KT-621 in non-Japanese healthy volunteers and patients with moderate to severe atopic dermatitis indicated rapid, deep, and sustained STAT6 degradation in blood and skin. These studies also showed robust reductions in disease-relevant Type 2 inflammatory biomarkers, meaningful improvements in clinical endpoints and patient-reported outcomes in atopic dermatitis, and positive results in patients with comorbid asthma and allergic rhinitis, all within a favorable safety profile.
